I think you missed to give the right permission.
Either ASPNET user permission is missed or the NETWORK SERVICE

That does sound odd. My first guess would have been the web service extensions which you said are good. Do static htm pages work?
A good tool is filemon from www.sysinternals.com. It's free and trustworthy on a production server. That will let you see if it's trying to do anything odd at the file system and if there are any file permission

I had that same type of issue a few weeks back. It was very odd. The way I fixed it was to run the "aspnet_regiis.exe -i" utility. Now I know it sounds funny that you would have to do that, but it made it work. Prior to running the tool, I was able to
see the "ASP.NET" tab for the properties of the IIS site. All looked well. I even switched it from 2.0 to 1.1 and then back to 2.0. Nothing worked. All that worked was running that tool.

If both ASP and ASP.NET pages are failing, then it's not something specific to just ASP.NET (or it's two separate issues). Are you sure it's a 404 error page you got? If you're getting a 403.1, then it's possible that the Execute Permissions are set to
None in your Home Directory tab. It should be Scripts Only.
The script mappings are in your Home Directory -> Configuration section. In there, make sure that you have .aspx, .asmx and .asp set to what they should be. Those would only have changed if someone changed them, but it's worth double checking.

Great, glad you got it going. It doesn't tell us why it failed in the first place, but it sounds like something in IIS was causing that, and not something with ASP.NET specifically, for example.

Sounds like a permission issue. Try using IUser and/or IWAM
