What is the best way to access database from asp.net application. I used application blocks before and also entity framework, but is there anything else better that I can use to access database from my application.

I would not use Microsoft.ApplicationBlocks.Data / Enterprise Library as to the best of my knowledge, they are not being maintained.
Depending on your requirements, I would consider using a micro ORM like
I use Entity Framework or Dapper when I'm building an application. If it's a very large application with complex mapping requirements - then I normally opt for a full ORM like Entity Framework. Otherwise, I stick with Dapper.
I may use straight ADO.NET if I am building a service or some utility that doesn't require any sort of mappings.

It is hard to give an correct answer to you. For which is based on your requirement.
Normally, we uses to use ADO.NET to connect database.
