I watched "Working with PNG-8" at
http://www.lynda.com/Photoshop-tutorials/Design-Web-Working-PNG-8/162712-2.html . Its about decreasing file size with PNG-8 without tangible loss of quality. I'm looking for a way of doing it programmatically. How can I do it with GDI+? there is no need

According to your description, i would suggest you check out the links for details:
PNG-8 or PNG-24
Convert 32 bit PNGs to high quality 8 bit PNGs
you have to perform some programming by your self.
