Our users are experiencing a noticible delay of the pop-up ajax toolkit calendar when they click on the date textbox. When we do it in our test environment it is much faster. Is there anything that can cause this or is there some fix? Thanks.

noticible delay of the pop-up ajax toolkit calendar
Most of the Ajax toolkit controls are clientside. So, calendarextender functionality depends on local computer resources and browser. If the client machine is slow for various reasons, you would notice the delay or unresponsiveness after textbox is clicked.

My suggestion applies when you notice memory leaks and page becomes unresponsive. For example, I use Chrome debugger tools to inspect memory leaks and investigate why UI is unresponsive. Here is the link I usually refer to
Let's say profiler shows 100MB as memory consumption after heap snapshot is taken, that's something to worry about. If your application is consuming 10MB memory, you may not have any issues. This matters if you open the page on PC having 256MB RAM compared
to pc having 16GB RAM. That's my two cents.
