What is the best way to archive single threaded environment
In asp.net MVC application.
I mean which one is most better and safer method?
Either Murex , Task or any other recommended approach.

You can try using a product such as NServiceBus(http://particular.net/) to offload the processing and run it in single threaded mode. The advantage to this is that all requests will be processed in order and the processing
can be offloaded from the web server as you don't really want long running processes to happen on a web server.

I think you are asking how to guarantee that only a single thread does 'something' at any point in time.
What is the 'something' that you want to protect? The reason there are many different ways to restrict to a single thread is because there are many different 'somthing' operations that people might want to do. There is no single answer (if there was, why
would all the other methods exist?)
So, what are you trying to do?
