Web Deployment Projects will not be supported in VS2012. This has always been an out-of-band add on which lagged behind the RTM release, but it won't be continued anymore. See
http://blogs.msdn.com/b/webdev/archive/2012/08/06/plans-regarding-website-projects-and-web-deployment-projects.aspx about why (hint: we're going to make Web Site publishing better without WDP).
As for the configuration manager - Web Site projects only support the Debug configuration. This is because technically, they don't even have a build configuration. Build configurations are an MSBuild concept and Web Site projects aren't based on MSBuild
(note: Web Application projects are MSBuild based). With WDP, which was a separate project and MSBuild based, it was able to support multiple configurations.

your explanation is confusing
Web Site projects only support the Debug configuration!!!!
so what should i do to have release mode?
what is WDP?

WDP is Web Deployment Project. It's the project type you added to deploy your website in the past.
There is no way to have a Release configuration for your website. Even if you could, since it's not an MSBuild based project, having a Debug or Release doesn't change how the project builds or runs. Those are MSBuild concepts and don't apply to Web Site
As the blog post I linked to mentions, we'll have a new release in the future which will provide the same functionality as WDP and more.
