I want to develop an iphone web application.
How can I do it using Visual Studio??

What do you mean by an iPhone web application? If you mean a website that can be used by an iPhone, it is just a normal site but you have to make sure your mark-up etc is compatible with the iPhone's browser.
If you mean an iPhone app, then you can't, you have to use Objective C and a Mac.

To develop mobile applications for iphone(or any other mobile) you can just use html and CSS. In case if you need much client side interaction you have to opt for mobile JQuery. And you can develop mobile web applications using small applications like KenduUI.

You have to use mobile JQuery and JSON results (for data). The server side concept is not there in mobile web applications instead there is a common standard JSON or xml for Data representation and transfer between client and server.
The server side code can be any webservice, wcf service or any other method to get the data.
Mobile browsers have limitations in terms of memory so generally we programmers will try to reduce the overload on the mobile devices so that device will not hang.
Still You can use asp.net to develop your web applications but there is no guarantee that all the functionalities will work on mobile. browsers.

You need a IPhone emulator. search the google and check whether any IPhone emulator is availble for windows or not. On Mac it's already avaialbe with Xcode.
