in a previous post (http://forums.asp.net/t/1794993.aspx/1?Using+a+custom+RouteHandler) I have created a httphandler which derives from the PageRouteHandler. I managed to fix some early problems, but now I have run into a new problem: RequestContext.Current.Session
I have read around a bit, and mostly people say all I need to do is derive my class from the instance System.Web.SessionState.IRequiresSessionState
This, however, does not appear to enable the session for me.
Any help in this would be greatly appreciated.

As stated in my post, I have already tried using the IRequiresSessionState interface, it did not work.
Could this be because IRequiresSessionState expects an IHttpHandler, while I am using an IRouteHandler?

Yes. Session state is not available to an IRouteHandler, it isn't created at that point yet.
