You can insert record from desktop application to web server DB, which can be achive by simply adding two connection string for both the databases.
But from web application to your local DB, you have to take a static IP address. After Static IP you can do it in the same way.

If you have same structure DB on site site then take backup of desktop DB and restore on other site.
If you want to copy record of some tables then create image DB of web application then copy all tables which you want to use on web application site. then take backup and restore to other site.
create table with record.
use this query,

Thanks for your quick replies. Do I need to create and use webservices once and they will be used for both website and desktop application? Will they be secure?

Is this database SQL Server? Before we jump to services (which might be the solution) I would like to know some additional information. Is the desktop version have some problem accessing the database directly? If it has access to a service it must have
connectivity. Can you give some detail as if this is distributed software (maybe packaged) that can not access the db, or is this software within a corporate domain possibly?
