The "Getting Started " and "Learn" sections of this site (asp.net) have many online tutorials, videos and walk-throughs. I would start there. You'll stay busy for days watching and learning! ;)

You best option might be to start with a 'Starter Kit' from this site; they are essentially complete applications that are generic enough to be modified for your specific needs. In fact, this site includes a starter kit for exactly what you are requesting:

Here is the full list of starter kits if you want to see them as well:

As far as the architecture goes, you can start by following the design of the starter kits to become familiar with a good way to build your project. Architecture in .NET is like anything else; you get better and improve your design each time you build another
You may want to read a few OOP books that delve into a basic n-tier (UI -> BLL -> DAL) architecture to begin. I have not read this entire book, but it does get into the design concepts that you may be looking to learn:

Thanks for all the responses
Actually, I have some experience building sites and web applications (almost did 15 sites and some web applications, so what I need to know is how to build web applications using advance techniques. Like using the tire and the layer architecture
and if I need to use design pattern too. In addition, I need to know how I can incorporate this architecture with wcf, jquery, json, linq and Client-Side AJAX . Therefore, I need to be able to take some online courses/training that'll allow me to get some
high level training without having to go through all the basics.
As always, your advice is greatly appreciated..
