I can get the following URL to work in IIS V7.0
web.config IIS V7.0
<add verb="*" path="wsDataAction" name="MyHandler" type="MyHandler"/>
I don't know how to get the same result in IIS 6.0. I tried this in the web.config file with bad results. IIS 6.0 seems to want a file extension on the path.
web.config IIS V6.0
<add verb="*" path="wsDataAction" type="MyHandler, MyHandler" />

I think the short answer is that you can't in IIS6 AFAIK only IIS7 supports this feature

That is IIS 7's feature that allows you to add mapping to managed handler. However, it looks like you are seeking for function like Url Mapping:
See if these are what you are looking for.
