<add name="MyConn" connectionString="Provider=Microsoft.Jet.OLEDB.4.0;Data Source=D:\Tirupathi\thirupathi.mdb;"
You can reference it via the Connections dropdown in the SqlDataSource control configuration wizard, or through code:
You can't use a connection string with an AccessDataSource control. Also, I would put the mdb file in the App_Code folder, then the string will always be:
which means that you won't have to change it when you copy the web site to another machine, such as a web server.

<add key="accessstring" value="D:/Tirupathi/thirupathi.mdb" />
Make sure that the database is in the Tirupathi folder on your D drive. Then, if you still can't get that to work, post the error message you are getting and the code that causes it.
