I have install this starter kit with SQL server 2005. I get the error in the bold line, and it say "The user instance login flag is not supported on this version of SQL Server. The connection will be closed". How to solve it?

We need more information as to what problem you are having.
Is it a local problem, or an online server problem?
What program do you use to edit your website?
Why are you showing code for checking the adexpiration?????
And instead of quoting the error copy and paste it so we can see what's happening.

I have install the sql server 2005, below is the step by step i configure :
1. Change Web.Config ConnectionString
2. Change the membership
When i run the login.aspx it has error at the line, which is under maintenance.vb. Anything else i need to change if i using sql server 2005?

You still didn't answer my question if the application works on your local computer??????
And I don't know if this is a local problem or a server problem
But asuming that it does work locally, and not online, it looks like your web.config is not pointing to both databases.
And it cannot find your members info, try something like this.
<add name="classifiedsConnection" connectionString="Data Source=xxxxxxxxxxxxxxxxxxxxxxxx;
<add name="LocalSqlServer" connectionString="Data Source=xxxxxxxxxxxxxxxxxxx;

I am using local database thats why my connectionstring will point to the local database
