My first rollout was on SQL 2000 but have since upgraded all sites I maintain to 2005. My memory was there were no initial problems with setting up the live site on 2000.
The only issues I recall were due to using SQL Express 2005 on the local environment and migrating data to the live SQL Server 2000. Some scripts I use to upload data had to change to accommodate small differences in tables structures between the two environments.
The moral is to keep the development environment as close as possible to the production environment i.e. i'd advise getting the starter kit working on your local PC using SQL Server 2000 rather than using the default SQL Express.
