I have just installed IIS manager version 6.1 build 7600 and visual studio 2010 (latest additions of both).
I have followed all the instructions and given the correct permisions ect for IIS to work with my directory. I have a helloworld website that has worked via the localhost server many times, but almost hourly something happens to the settings in IIS and it
starts throwing errors.
The current error copied below. Basically I am wondering a few things...
1. Why does my IIS constantly do something so that it works one minute and then it wont the next (this is with an identical helloworld.htm file so I know it is not something I am doing with the file that is messing it up).
2. How do I fix this error.

Have a look at this thread http://forums.asp.net/t/983377.aspx. Looks like you may have 64-bit only IIS trying to load 32-bit ASP.NET application.

Thank you for your answer, unfortunatly that was not the solution I was looking for.
I checked this forum before and was unable to find any solution.
The only thing I can think of is that I instaled Dungeons and Dragons online and that came with .net 1.1 so maybe installing that has messed up my settings somehow?

Quite possible - it could change default App Pool to 1.1 in IIS. Try running aspnet_regiis -i from command prompt in 2.0 or 4.0 folder.

How do I do that?
Forgive me, I just started using .net and am still at a very basic level.

Open command line prompt as administrator, go to Windows\Microsoft.NET\Framework\v2.xxxx, run aspnet_regiis -i. You can also use IIS control panel in Computer | Manage to set default ASP.NET version.

Thank you very much. It now works, although some more settings need to be tweaked but the basic problem was solved.
