  You need to register your WebUserControl in the master page with placing the following at the beginning of the file. <%@ Register src="regions.ascx" tagname="regions" tagprefix="uc1" %> Then you could use it in the master. But normally you leave the contentplaceholder empty, only when you like having default content for some content pages.
